This page is meant to keep the current configurations of all the 2-photon microscopes we have in an easy to access way. Information useful for maintenance or troubleshooting, like serial numbers, who used them in the past, RMA numbers, and when components were modified. Try to maintain all sections as you make changes, add in notes if something was repaired or is not functioning properly. Try not to add sections that are under flux unless necessary.
Microscopes are named Berg I through IV, they are all off-the-shelf from Thorlabs and run ScanImage Premium (see /two-photon-imaging/scanimage-setup-download-installation/).
Berg I#
Summary: Located at the ‘back’ of the longer air table with the side-facing light-enclosure opening. It is a galvo-res only system using a tunable laser for imaging and upgraded PMTs. It has NIDAQ-based acquisition capable of 2-channel acquisition.

Imaging Scanhead + Driver: Thorlabs Galvo-Res and Thorlabs ECU
- This was replaced using the Galvo-Res previously on Berg-II after notable damage to the original mirrors was found. Scanhead driver is NOT matched exactly to the mirrors, but it has not caused any issues so far.
- This is an older model that does not have galvo (mirror) position feedback, so you cannot calibrate the slow mirror, or optimize the scanning waveforms.
- The serial for this is the same as the system: TP0117080-1760
Imaging Laser + Modulation: Coherent Vision-S with Conoptics 350-80LA pockels cell
- This table has a single tunable femtosecond laser split equally between both microscopes
- Modulation by Conoptics pockels cell within a thorlabs BCM-PCA enclosure
- Feedback from a photodiode monitoring Pockels cell output sent to ScanImage
- Pockels Cell Amplifier Model 302RM is on the shelf above the air table, Serial:
- Note: Conoptics amplifier SN 3605 was repaired in 2018 (not yet sure if is on Berg I or II)
Fast-Z: Physik Instrumente PiFoc Piezoelectric Z-scanner + Piezo Driver
- Z-scanner part number: P-725K085, serial: 115066019
- Piezo driver part number: E-709.CHG, serial: 11567108
- HMS Asseet tag: HMS52041550
- Feedback about position from the piezoelectric actuator is sent to scanimage, so waveforms for z-stacks can be optimized.
Objective: 20X Olympus XLUMPLFLN Objective, 1.00 NA, 2.0 mm WD
- HMS asset tag: HMS36462
- Serial:
Detection: Thorlabs PMT2100 GaAsP PMTs
- These were upgraded in 2023 (courtesy of Pablo) to the Thorlabs USB-controled/powered models
- Originally had Hamamatsu H10770PA-40 with Thorlabs transimpedance amplifier TIA60
Acquisition: National Instruments NIDAQ PXIe-1073 Chassis System with cards
- PMT signals digitized (acquired) on NI 5732
Stage/Objective XY/Z Motor:
- Unclear if it is the original from install or if was repaired in the years since.
